چکیده

BACKGROUND In vitro maturation (IVM) of human oocytes is an emerging procedure quickly incorporated into the world of assisted reproductive technologies. As an effective method of in vitro maturation, several studies have reported the critical role of differentions on activating the complex process involved in both gamete maturation and fertilization. OBJECTIVE In this study, we supplemented a chemically defined medium with different combinations of selenium, calcium and calcium ionophore concentrations to obtain the best rate of human oocytes maturation, survival, and fertilization. MATERIALS AND METHODS As an experimental study, Three combinations of [selenium (5 μg/ml), calcium (5 μg/ml) and calcium ionophore (1 μg/ml)], [selenium (10 μg/ml), calcium (7 μg/ml) and calcium ionophore (2 μg/ml)] and [selenium (15 μg/ml), calcium (10 μg/ml) and calcium ionophore (5 μg/ml)] added to the chemically defined medium and the morphology of oocytes assessed after 22-24 hours in vitro maturation of the oocytes. RESULTS The highest percentage of MII (meiosis II) oocytes (68%), developing beyond the morula (20.1%) and the blastocyst formation (11.1%) observed in oocytes treated with 15µg/ml selenium, 10µg/ml calcium and 5µg/ml calcium ionophore. Moreover, we showed the significant rate of survival in each three combinations after 36, 72 and 96 hours. CONCLUSION Maturation and activation of oocytes may be triggered by changes in intracellular ion concentrations as second messengers in signal transduction pathways. Here, we received the highest percentage of in vitro maturation and fertilization among three combinations of selenium, calcium and calcium ionophore treatments. Using this combination of ions beside other factors might be useful for the enrichment of the human oocytes IVM medium.

P-37: The Effect of Extracellular Calcium Concentration on the In vitro Maturation of Mouse Oocytes after Mechanical Stimulation by Hydrostatic Pressure

Background: Oocyte maturation is usually defined as the period of progression from the first to the second meiotic arrest and involves coordinated nuclear and cytoplasmic modification. Ca2+ signals are known as important regulators of oocyte maturation. Extracellular Ca2+ is important for first polar body formation, normal meiosis kinetics, and for preserving the chromosome and spindle configur...

Background: Despite great advances achieved in the field of in vitro oocyte maturation (IVM), almost all IVM media are supplemented with undefined component which not only pose the risk of pathogen transmission but also hinders our knowledge of molecular mechanisms of maturation. One of these components is serum that containing various concentration of unknown molecules affecting cellular proce...

Effects of different activation protocols on cleavage rate and blastocyst production of caprine oocytes

The present study was undertaken to assess the effect of different chemical activators along with 6-DMAP on in vitro matured caprine oocytes. From 4332 ovaries, 14235 cumulus oocyte complexes (COCs) were collected which were matured in TCM-199 medium containing follicle stimulating hormone (FSH) (5 µg/ml), Leutinizing hormone (LH) (10 µg/ml), oestradiol-17β (1 µg/ml) supplemented with 10% fetal...

P-76: Cytogenetic Investigation of Parthenogenetic Mouse Embryos Generated from In Vitro Activated Oocytes by Hydrostatic Pressure in The Presence of Calcium Ionophore and Ethanol

Background: The advances in cytogenetic techniques during the last few years have permitted not only the study of large populations of wild and domestic animals, but also the detection of chromosome anomalies in embryos. Chromosomal abnormalities are the most common cause of embryonic and fetal mortality in mammals. Most reports of chromosome anomalies in parthenogenetic embryos describe numeri...

P-94: The Effect of Calcium Ionophore A23187 and Ethanol on Parthenogenetic Activation of Mouse Oocytes in Presence of Hydrostatic Pressure and Cy-tochalasin B

Background: Parthenogenetic activation of mammalian oocytes using artificial stimuli is commonly used in various reproductive bio-techniques. Calcium ionophore is known to elevate intracellular calcium levels in the cytoplasm of oocytes through the influx of calcium from extracellular spaces. Ethanol promotes a single intracellular Ca2+ increase of greater and longer amplitude than the initial ...
